Enhancement Dissolution of Celecoxib by Surface Solid Dispersion with Avicel PH 101 as A Matrix

Abstract
Surface solid dispersions (SSD) system could increase the solubility and dissolution of drugs that are poorly soluble in water like celecoxib. This study aims to determine the effect of the formation of SSD system with Avicel PH 101 as a carrier on the dissolution of celecoxib. The SSD of celecoxib was prepared by dissolving method using Avicel PH 101 as a carrier in a ratio of 1:1, 1:2 and 1:3. SSD system did dissolution test with buffer pH 12 (0,04 M tribasic sodium phosphate) and 1% SLS. SSD system did X-ray diffraction for determinate crystal habit. The results showed that the SSD celecoxib with Avicel PH 101 carrier of all ratios was able to increase the dissolution of celecoxib compared to pure celecoxib and the physical mixture. The DE60 value of SSD celecoxib ratio 1:1 was 57.93 ± 1.53; 1:2 ratio of 59.86 ± 0.53; a 1:3 ratio of 63.94 ± 0.40. SSD system characterized by sloping diffractogram curve. There was a significant difference between the dissolution of the SSD system, the physical mixture and the pure celecoxib. SSD system could change crystalline habit of celecoxib was be amorphous.
