Bioactivity-Guided Isolation of Antioxidant Compounds from Premna serratifolia L. Leaves Using the DPPH Assay

Abstract


Premna serratifolia L., locally known as bebuas, is one of the native plants of Indonesia that has long been used as a traditional medicine, especially in the Kalimantan region. In this study, an evaluation of the antioxidant activity of P. serratifolia leaf extract and fractions was conducted to identify bioactive compounds that have the potential as antioxidants. This study aims to isolate active compounds from bebuas leaf extract with a bioactivity approach guided by the DPPH (2,2-diphenyl-1-picrylhydrazyl) radical inhibition test. Isolation of compounds was carried out in stages starting from the ethyl acetate fraction, which showed the highest antioxidant activity based on the initial test results. The fractionation process was carried out using vacuum liquid chromatography (VLC), then continued with purification using permeation gel column chromatography with Sephadex LH-20. The purified subfractions were further separated using preparative thin layer chromatography (TLC) until three pure compounds were identified as flavonoids. The characterization of the three compounds was carried out through a combination of methods, namely thin layer chromatography with specific spot detectors, UV-Vis spectrophotometry, and mass spectrometry. The results of the analysis showed that the compounds obtained were flavonoids, which contributed to antioxidant activity in the ethyl acetate fraction of P. serratifolia leaves.
